Abstract
B-Cell Navigator is a dual-engine platform for rapid, scalable discovery of target-specific antibodies at single-B-cell resolution, from the full immune repertoire. The two engines interrogate distinct B-cell compartments and are typically run in parallel — or independently when target format or screening requirement dictates. Engine 1 uses imaging-based spectral sorting to interrogate the memory B-cell compartment, with per-cell visual confirmation of antigen binding. Engine 2 uses droplet-based microfluidic screening of antibody-secreting cells, selecting on binding characteristics or functional activity directly.
Qualified hit panels are delivered 8–10 weeks post-immunisation, ready to enter conventional lead discovery and optimisation. Alternatively, natively paired VH:VL sequences can be recovered at scale and transferred into our mammalian display system, enabling CMC-quality lead selection in the final therapeutic format (mono- or bi-specific IgG) and de-risking developability before production commitment. While the data shown here are from huOX40 immunised mice, the platform is equally deployed on B cells isolated from PBMCs of human disease donors
