Workflow

01

Stable pool generation

Synthetic genes or sgRNAs are designed, ordered and cloned in appropriate plasmids from an in-house library of over 50 vector backbones.

Transfection and initial selection take place to generate the stable pools Expression or functionality is tested both 24h after transfection and at stable pool stage.

Pools can be used directly for antibody library selections and early-stage screening where single-cell clone homogeneity is not yet required.

Single-cell clone selection

Single-cell FACSorting to obtain desired expression level (e.g. low and high-expressing clones).

Up to 30 clones (overexpression) or 60 clones (reporter/knockout) are screened by expression levels or functional performance.

Single-cell clone final characterization

Freeze-thaw viability tested. Target overexpression levels confirmed by flow cytometry.

Reporter cell lines validated for signal induction and fold-change upon stimulation and EC50/IC50 calculated for reference antibodies.

Both are tested for stability expression and performance over 2-weeks. Knockout cell lines confirmed by sequencing and target loss-of-expression.

Timeline

Single-cell clone stable cell lines delivered within 3-4 months. Pools available in 6-8 weeks. Specific timelines confirmed at project initiation based on target, format, and host cell line.

  • Cold storage and BioLogistics: cells can be shipped to the partner or stored at −196°C (liquid nitrogen) at FairJourney Bio
